ABSTRACT

Objective: To determine and compare the remineralizing capacity of self-assembling peptide (SAP) P11-4 and casein phosphopeptides­amorphous calcium phosphate (CPP-ACP) on enamel. Material and Methods: Enamel samples were divided into 2 groups. Group I was treated with Self­assembling peptide (SAP) P11-4 and group II with casein phosphopeptides-amorphous calcium phosphate (CPP-ACP). In both groups, remineralizing capacity was assessed at baseline, 2 weeks, 6 weeks and 12 weeks. Student's t- test and ANOVA were applied, with the significance level set at 5%. Results: The mean calcium weight % was evaluated at baseline, 2 weeks, 6 weeks and 12 weeks. In Group I, there was increase in mean value (62.12 ± 1.24) from baseline to 12 weeks (67.36 ± 2.14). However, there was decrease in phosphate weight % from 37.16 ± 2.52 at baseline to 35.72 ± 2.11 at 12 weeks. In Group II, mean calcium weight % was 64.18 ± 1.52 at baseline, which ultimately increased to 66.01 ± 2.03 at 12 weeks. Phosphate weight % showed reduction from 37.34 ± 2.23 at baseline to 35.04 ± 2.02 at 12 weeks. Ca/P ratio showed significant improvement. There was significant difference in Ca/P ratio at 2 weeks, 6 weeks and 12 weeks in both groups (p<0.05). Conclusion: Self-assembling peptide (SAP) P11-4 found to be more effective and efficient as compared to casein phosphopeptides­amorphous calcium phosphate (CPP-ACP).

ABSTRACT

Objective: To evaluate and compare the remineralization potential of a dentifrice containing bioactive glass and a topical cream containing casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) in remineralizing artificial carious lesion on enamel. Material and Methods: Forty-five freshly extracted human permanent premolar teeth were selected. Samples were divided into three groups: GI - regular tooth paste without specific remineralizing agent; GII - tooth paste containing calcium sodium-phosphosilicate (novamin) and GIII - topical cream containing casein phosphopeptide-amorphous calcium phosphate. All the sound enamel samples were viewed under scanning electron microscope (SEM) to assess the topographical pictures of enamel surface and energy dispersing x-ray analysis (EDAX) was done to estimate quantitatively the amounts of mineral (calcium and phosphorous). The mineral content of calcium and phosphorus after demineralization in each group was noted. The samples were then subjected to SEM and EDAX. Results: GI does not show any increase in the calcium and phosphorus after applying toothpaste without any remineralizing agent but GII and GIII showed a net increase in calcium and phosphorous values after applying concern-remineralizing agents. Inter group comparison showed GIII yield higher net calcium and phosphorous values than GII. Conclusion: Two remineralizing agents showed remineralization potential on enamel surfaces. Casein phosphopeptide-amorphous calcium phosphate showed better remineralizing potential than calcium sodium phosphosilicate. Hence CPP-ACP can be considered as the material of choice in remineralizing early enamel carious lesions.

ABSTRACT

Objectives: To investigate the effect of different mixtures from nano casein phosphopeptides (CPP), amorphous calcium phosphate (ACP), probiotic Lactobacillus rhamnosus B-445 (L. rhamnosus) and casein glycomacropeptide (GMP) against Streptococcus mutans (S. mutans) growth and its adhesion. Methods: CPP was prepared from the tryptic digest of bovine casein and GMP by the action of chymosin on casein solution. Four mixtures namely: Group 1) Nano CPP; Group 2) Nano CPP+ ACP; Group 3)Nano CPP+ ACP + L. rhamnosus; and Group 4)Nano CPP+ ACP+ GMP were prepared and tested for its inhibitory activity against S. mutans growth and its adhesion to saliva-treated glass surfaces in comparison with a commercial product (GC MI paste plus) and chlorhexidine (0.2%) as a positive control.  The particle size and zeta potential of nano CPP and its complex with ACP were evaluated. Furthermore, the viability of L. rhamnosusin its mixture was determined during two weeks of storage at pH 6.8 and 8 respectively. Results: Revealed that Nano CPP had an average particle size (7.75 nm) and zeta potential (-8.43 mV) lower than that of CPP+ACP mixture. Probiotic containing mixture exhibited inhibitory activity slightly less than the positive control at pH 6.8. All tested mixtures reduced the adhesion of S. mutans to saliva-treated glass surfaces and the highest was that containing probiotic and GMP. L. rhamnosus showed acceptable stability in CPP+ACP mixture during storage period. Conclusions: All these findings suggest the use of probiotic, CPP+ACP mixture as a dental anticariogenic and remineralizing agents.

ABSTRACT

Muitos métodos têm sido empregados na produção de peptídeos bioativos para a promoção da saúde. O objetivo deste estudo foi produzir caseinofosfopeptídeos por hidrólise tríptica do caseinato de sódio usando diferentes temperaturas (37 e 50 °C) e tempos de reação (2 e 4 h), caracterizá-los e analisar sua influência nas atividades citotóxica, antimicrobiana e antioxidante. Caseinofosfopeptídeos foram caracterizados através da composição centesimal, eletroforese em gel de poliacrilamida, Espectrometria de Massas e Cromatografia Líquida de Alta Eficiência. Toxicidade para leucócitos humanos, atividade antimicrobiana utilizando o teste de microdiluição em caldo e determinação da capacidade antioxidante pelo método de espécies reativas ao ácido tiobarbitúrico foram os ensaios biológicos realizados. Os resultados mostraram que as quatro frações peptídicas obtidas apresentaram-se com baixo peso molecular e elevados teores proteico e mineral; quanto ao perfil aminoacídico, apresentaram elevadas e diferenciadas quantidades de ácido glutâmico e serina, que pouco variaram de acordo com o processo de obtenção; não se mostraram tóxicos para leucócitos humanos; demonstraram atividade antimicrobiana para Escherichia coli e Salmonella Enteritidis e elevada capacidade antioxidante. Os resultados físico-químicos das frações de caseinofosfopeptídeos demonstraram elevada composição nutricional em termos de proteína e, principalmente, cálcio. O conjunto de dados indicou que alterações no tempo e na temperatura de reação para a obtenção dos hidrolisados não interferem nas suas qualidades biológicas, mostrando serem seguros para a promoção da saúde e para a aplicação em situações especiais, que envolvem pacientes desnutridos, imunossuprimidos, com comprometimento ósseo ou gastrintestinal decorrentes de inflamações e infecções.


Several methods have been employed for the production of bioactive peptides for health promotion. The aim of this study was to produce and characterize Casein phosphopeptides obtained by sodium caseinate tryptic hydrolysis under different temperatures (37 and 50 °C) and reaction times (2 and 4 h), and evaluate their biological capabilities. They have been characterized by assessing their centesimal composition, by polyacrylamide gel electrophoresis, Mass Spectrometry, and High-Performance Liquid Chromatography. The biological activities tested included toxicity for human leukocytes, antimicrobial assay using the microdilution test, and determination of the antioxidant capacity by the thiobarbituric acid reactive species method. The results showed that the four fractions obtained were of low molecular weight with high protein and mineral contents; their amino acid profile showed high and differentiated amounts of glutamic acid and serine independent of the methodological procedures. The results also showed no toxicity for human peripheral leukocytes, demonstrated antimicrobial activity against Escherichia coli and Salmonella Enteritidis as well as high antioxidant capacity. The results of the physicochemical Casein phosphopeptides' fractions showed high nutritional composition in terms of protein and, particularly, calcium. The biological assays indicated that time and temperature changes in the process for obtaining casein hydrolysates have not interfered with their biological qualities. In addition, they have proven safe in promoting health in special conditions involving malnourished and/or, immunocompromised patients or those with bone and/or gastrointestinal impairment due to inflammations and infections.

ABSTRACT

OBJECTIVE: A literature review of systematic reviews and in situ and in vivo randomized controlled trials was conducted in order to assess the role of casein phosphopeptide - amorphous calcium phosphate in providing caries-preventive effect superior to any intervention or placebo. METHODS: Initially, Pubmed database was searched for systematic reviews. Two systematic reviews were identified, which concluded that the quantity and quality of randomized controlled trials were insufficient to recommend the clinical use of the product. After this, Pubmed database was searched for in situ and in vivo randomized controlled trials that were not included in the reviews identified. In vivo and in situ studies yielded mixed results. RESULTS: In situ studies indicated greater efficacy of casein phosphopeptide - amorphous calcium phosphate in comparison with placebo. Whereas, in vivo studies demonstrated different findings CONCLUSION: Casein phosphopeptide - amorphous calcium phosphate had a performance equivalent to or greater efficacy than placebo; fluoride was more effective than the casein complex; casein phosphopeptide - amorphous calcium phosphate associated with fluoride showed better or equivalent performance to that of placebo. In view of available evidence, it was not possible to generalize the indication of casein phosphopeptide - amorphous calcium phosphate for preventing dental caries. OBJETIVO: Foi conduzida revisão da literatura em busca de ensaios clínicos randomizados in situ e in vivo que avaliassem o papel do complexo de fosfopeptídeo de caseína - fosfato de cálcio amorfo na prevenção de lesões de cárie. MÉTODOS: Inicialmente, buscou-se identificar na base Pubmed revisões sistemáticas sobre o tema. Foram identificadas duas revisões sistemáticas, que concluíram que a quantidade e qualidade dos ensaios clínicos randomizados são insuficientes para recomendação do uso clínico do produto. Posteriormente, pesquisaram-se ensaios clínicos randomizados in situ e in vivo que não tivessem sido incluídos nas revisões identificadas. RESULTADOS: Os estudos in vivo e in situ tiveram resultados contraditórios. Os estudos in situ indicaram uma maior eficácia do complexo de fosfopeptídeo de caseína - fosfato de cálcio amorfo em relação ao placebo. Já os estudos in vivo demonstraram diferentes. CONCLUSÃO: Complexo de fosfopeptídeo de caseína - fosfato de cálcio amorfo apresentou desempenho ora superior, ora equivalente ao placebo; o flúor demonstrou maior eficácia que o complexo de caseína; o complexo de fosfopeptídeo de caseína - fosfato de cálcio amorfo associado ao flúor se mostrou ora melhor, ora equivalente ao placebo.
